A Breakthrough in Future Medicine: NSCLC Diagnostic Technology Combining AI and CRISPR
Lung cancer is a fatal disease causing over two million deaths worldwide annually, with non-small cell lung cancer (NSCLC) accounting for more than 85% of all cases. While early diagnosis of NSCLC can significantly increase survival rates, current methods such as CT scans and biopsies are expensive, invasive, and limited in accuracy, creating an urgent need for the development of new biomarkers. Consequently, recent research has highlighted microRNA (miRNA) as a promising biomarker for NSCLC diagnosis. miRNAs are small RNA molecules consisting of only 20 to 25 nucleotides that regulate the expression of cancer cells. However, their practical application has been hindered by their extremely low abundance in blood and tissues, as well as high inter-individual variability.
To address these challenges, this study developed a CRISPR/Cas13a system that integrates artificial intelligence (machine learning) with hydrogels. CRISPR/Cas13a is a gene-editing tool targeting RNA, capable of recognizing, amplifying, and detecting specific miRNAs. By encapsulating this system within a polymer hydrogel, the research team increased miRNA sensitivity by more than 100-fold. Furthermore, using machine learning algorithms to analyze miRNA expression patterns in NSCLC patients versus healthy individuals, the team discovered 15 new biomarkers. Among these, miR-1246, miR-3195, and miR-4732 demonstrated the highest accuracy for NSCLC diagnosis, achieving a diagnostic accuracy of 92.3% using these three miRNAs alone.
Crucially, this technology enables real-time analysis. While conventional PCR-based miRNA detection takes several hours to over a day, the CRISPR/Cas13a system can produce results within just three hours. The research team predicted that combining this system with computed tomography (CT) could improve the early diagnosis rate of NSCLC by more than 25%. This is expected to be particularly useful for patients in the early stages of NSCLC. Currently, the five-year survival rate for NSCLC is over 60% when diagnosed early, but drops precipitously to below 5% when diagnosed in middle or late stages. Therefore, the practical implementation of this technology is expected to contribute significantly to improving survival rates.
The research team also verified the safety of the system. Because the hydrogel is highly biocompatible and biodegradable, it caused almost no side effects when injected into blood or interstitial fluid. Additionally, since CRISPR/Cas13a was used for detection rather than gene editing, there is no risk of genetic modification. The team stated, "This technology can be applied to discover biomarkers for other types of cancer or chronic diseases beyond NSCLC," and they plan to commercialize it through future clinical trials.
