Mitochondria within muscle cells serve as the powerhouses of energy production. As research into how exercise regulates these organelles intensifies, a new finding has emerged: small molecules called microRNAs (miRNAs) form a distinct regulatory network inside mitochondria.
This study compared mitochondrial miRNA profiles in male skeletal muscle before and after exercise. Researchers obtained muscle biopsies from healthy adult men, isolated mitochondria, and performed miRNA sequencing.
A distinct population of miRNAs existed within skeletal muscle mitochondria, distinguishable from cytosolic miRNAs. Following exercise, the expression of specific mitochondrial miRNAs changed significantly, and these miRNAs had target genes associated with mitochondrial gene expression regulation, oxidative phosphorylation, and mitochondrial biogenesis pathways. This evidence suggests that mitochondria are not merely passive energy converters but actively participate in exercise adaptation through their own gene regulation system.
These findings indicate that mitochondrial miRNAs may represent a novel molecular mechanism of exercise adaptation. Limitations include the male-only sample precluding sex-based analysis and the early stage of functional validation for these miRNAs.
While direct practical guidance for athletes remains premature at this basic research stage, mitochondrial miRNA profiles may eventually serve as biomarkers of exercise adaptation. The existing evidence that regular aerobic exercise promotes mitochondrial function and biogenesis remains valid. At least three sessions per week of 30-plus minutes of moderate-intensity aerobic exercise is the baseline for mitochondrial health.
