You may have experienced a feeling of chest tightness while breathing or noticed that your recovery after exercise is slower than usual. The phenomenon where body tissues lose their flexibility and harden may not be a simple process of aging, but rather dependent on how specific nutrients are metabolized. In particular, it has been revealed that certain amino acids, long known to be essential for muscle growth, can sometimes contribute to organ damage.
Published in the international journal JCI Insight, this observational and experimental study examines cellular changes occurring during the process of pulmonary fibrosis. Using human lung fibroblasts and precision-cut lung tissue models, the research team tracked the impact of alanine, a type of amino acid, on tissue transformation.
The study confirmed that alanine, one of the primary components of skeletal muscle, acts as a key fuel driving the deformation of lung tissue. The results showed that Transforming Growth Factor-beta (TGF-β), a signaling molecule in the body, drastically increases intracellular alanine levels. This occurs because the cells simultaneously increase their internal production of alanine and expand the pathways used to import alanine from external sources.
Of particular note is the role of glutamate-pyruvate transaminase 2 (GPT2), which drives alanine synthesis. When the activity of this enzyme was inhibited, intracellular alanine levels dropped, and the fibrotic reaction—the hardening of the tissue—was significantly reduced. Conversely, when alanine was artificially supplemented from an external source, the halted fibrotic process was observed to accelerate once again.
Alanine serves not merely as an energy source but as a regulator that determines the overall metabolic flow of the cell. According to the research, alanine is essential for maintaining glycolysis and the citric acid cycle, the body's primary energy-generating circuit. Furthermore, alanine supported abnormal structural changes in the tissue by providing the carbon and nitrogen necessary for the production of proline and glutamate, the primary raw materials for collagen synthesis.
The researchers also identified the role of the amino acid transporter family member 38 member 2 (SLC38A2), which transports alanine. When cells perceive a deficiency in alanine, they increase the expression of this transporter to absorb more alanine and glutamine from the environment. Consequently, the most effective way to prevent abnormal tissue hypertrophy and fibrosis was to block both the synthesis and import pathways of alanine simultaneously.
This study suggests that alanine, commonly consumed in exercise supplements, can accelerate pathological tissue changes under specific disease conditions. While alanine has traditionally been viewed solely as a beneficial amino acid that aids in muscle fatigue recovery, this research reveals it as a core element of metabolic reprogramming that alters cellular morphology.
However, since this study was conducted on cells in the specific disease state of pulmonary fibrosis, it is difficult to conclude that alanine intake in a generally healthy state directly causes tissue fibrosis. Rather, the study holds significant value as foundational research demonstrating how imbalances in protein metabolism can trigger chain reactions affecting organ health.
To maintain healthy tissue flexibility in daily life, a balanced intake of amino acids is paramount. It is recommended to obtain amino acids through high-quality natural proteins such as poultry, fish, and legumes, while adjusting the use of highly concentrated supplements according to one's individual health status.
Additionally, at least 30 minutes of regular aerobic exercise per day helps ensure smooth oxygen supply to lung tissues, which can help suppress abnormal metabolic changes. Incorporating green and yellow vegetables, such as spinach and broccoli—which are rich in B vitamins that support alanine metabolism—into the diet can be a practical way to promote healthy tissue recovery.
